Loops are good for routes that have a lot of variety. Loops have the advantage of having the trains run into each station more frequently because it doesn't need to go backwards. I find having lines end in squares on both sides to be very effective. Lines handle the late game better than loops especially when the special symbols start to pile up because they pick up only the passengers going that direction whereas loops pick up everything. Lines are good if you have a lot of circles back to back because then the train will pick up all the stuff on the way forwards and on the way back, whereas a one directional loop will only pick up all the passengers from the first circle and end up not having capacity for the second one.
